ABSTRACT

ObjectiveTo investigate the protective effect of the extract of Liuwei Dihuangwan (LW) on mitochondrial damage in the Alzheimer's disease (AD) model of Caenorhabditis elegans (C. elegans). MethodC. elegans transfected with human β-amyloid protein (Aβ) 1-42 gene was used as an AD model. The rats were divided into blank group, model group, metformin group (50 mmol·L-1), and low, medium, and high dose (1.04, 2.08, 4.16 g·kg-1) LW groups. Behavioral methods were used to observe the sensitivity of 5-hydroxytryptamine (5-HT) in nematodes. Western blot was used to detect the expression of Aβ in nematodes. Total ATP content in nematodes was detected by the adenine nucleoside triphosphate (ATP) kit, and mitochondrial membrane potential was detected by the JC-1 method. In addition, the mRNA expression of Aβ expression gene (Amy-1), superoxide dismutase-1 (SOD-1), mitochondrial transcription factor A homologous gene-5 (HMG-5), mitochondrial power-associated protein 1 (DRP1), and mitochondrial mitoprotein 1 (FIS1) was detected by real-time fluorescence quantitative polymerase chain reaction (RT-PCR). ResultThe extract of LW could reduce the hypersensitivity of the AD model of nematodes to exogenous 5-HT (P<0.05) and delay the AD-like pathological characteristics of hypersensitivity to exogenous 5-HT caused by toxicity from overexpression of Aβ in neurons of the AD model of nematodes. Compared with the blank group, in the model group, the mRNA expression of Aβ protein and Amy-1 increased (P<0.01), and the mRNA expression of SOD-1 and HMG-5 decreased (P<0.01). The mRNA expression of DRP1 and FIS1 increased (P<0.01), and the level of mitochondrial membrane potential decreased (P<0.05). The content of ATP decreased (P<0.01). Compared with the model group, in the positive medicine group and medium and high dose LW groups, the mRNA expression of Aβ protein and Amy-1 decreased (P<0.05,P<0.01), and the mRNA expression of SOD-1 and HMG-5 increased (P<0.01). The mRNA expression of DRP1 decreased (P<0.05,P<0.01), and that of FIS1 decreased (P<0.01). The level of mitochondrial membrane potential increased (P<0.01), and the content of ATP increased (P<0.05,P<0.01). ConclusionThe extract of LW may enhance the antioxidant ability of mitochondria, protect mitochondrial DNA, reduce the fragmentation of mitochondrial division, repair the damaged mitochondria, adjust the mitochondrial membrane potential, restore the level of neuronal ATP, and reduce the neuronal damage caused by Aβ deposition.

ABSTRACT

The fungus Xylaria sp. KYJ-15 was isolated from Illigera celebica. Based on the one strain many compounds (OSMAC) strategy, the strain was fermented on potato and rice solid media, respectively. As a result, two novel steroids, xylarsteroids A (1) and B (2), which are the first examples of C28-steroid with an unusual β- and γ-lactone ring, respectively, along with two new dihydroisocoumarin glycosides, xylarglycosides A (3) and B (4), were identified. Their structures were elucidated by spectroscopic methods, X-ray diffraction and electronic circular dichroism (ECD) experiments. All isolated compounds were evaluated for cytotoxicity, DPPH radical scavenging activity, acetylcholinesterase inhibitory and antimicrobial effect. Compound 1 exhibited potent AChE inhibitory activity with an IC50 value of 2.61 ± 0.05 μmol·L-1. The β-lactone ring unit of 1 is critical for its AChE inhibitory activity. The finding was further confirmed through exploring the interaction of 1 with AChE by molecular docking. In addition, both compounds 1 and 2 exhibited obvious antibacterial activity against Bacillus subtilis with a minimum inhibitory concentration (MIC) of 2 μg·mL-1. Compounds 3 and 4 exhibited antibacterial activities against Staphylococcus aureus with MICs of 4 and 2 μg·mL-1, respectively, which also exhibited DPPH radical scavenging activity comparable to the positive control with IC50 values of 9.2 ± 0.03 and 13.3 ± 0.01 μmol·L-1, respectively.

ABSTRACT

Objective:To analyze the advantages of IPSA combined with increasing cervical center dose in intracavitary and interstitial brachytherapy (IC/IS) for locally advanced cervical cancer.Methods:A total of 46 stage Ⅱ B cervical cancer patients with, local lesion size≥5 cm after 45 Gy/25f external intensity-modulated radiotherapy (IMRT) were recruited. Uterine tandem and needles were implanted, CT was performed, and then HR-CTV, rectum, bladder, sigmoid colon and the area of cervix increased dose (HR-cervix) were delineated, IPSA was used for optimization. According to whether the dose of HR-cervix was increased or not, all patients were divided into IC/IS+ HR-cervix group (group A) and IC/IS group (group B). The differences in dosimetric parameters were compared between two groups. Results:The relative uterine tandem dwell time was significantly extended in group A ( P<0.001). In group B, the V 150% and V 200% volumes of HR-cervix were increased from 63.94% and 30.80% to 91.54% and 64.06%. The D 90% and D 100% in group A were significantly lower than those in group B (both P<0.05). There was no statistical difference in organ at risk (OAR) dose. Conclusion:IPSA combined with increasing cervical center dose can meet the HR-CTV D 90% dose requirement, normal tissue dose limits, and can escalate the doses to local areas of the cervix.

ABSTRACT

Objective To investigate the clinical effect of improved portable mirabilite bag by abdominal application in the treatment of severe acute pancreatitis. Methods A total of 50 patients with severe acute pancreatitis admitted to our hospital from May2015 to January 2016 were enrolled as the control group, which was treated with traditional self-made towel bag filled with mirabilite for the abdominal application. Another 50 patients with severe acute pancreas hospitalized in our hospital from February 2016 to April 201750 patients as the experimental group, which was treated with improved portable mirabilite bag by abdominal application. The two groups were compared in terms of contamination by clothing, shedding, dislocation, aggregation, abdominal pain, bloating, and recovery time of gastrointestinal function. Results The incidences of shedding and displacement of mirabilite bag, mirabilite accumulation and contamination in the experimental group were lower than those of the control group. The abdominal distension time and bowel sound recovery time were significantly shorter than that in the control group (P <0.05, P <0.001). Conclusions By the improved portable mirabilite bag, the mirabilite can be evenly applied to the external abdomen of the patients. It is simple to make the bag and convenient for the clinical use, worthy of clinical application.

ABSTRACT

Objective To investigate the dosimetric difference between inverse planning simulated annealing(IPSA)and manual optimized plan for isodose line in interstitial brachytherapy for locally advanced cervical cancer and to provide a better optimization method for clinical application. Methods A total of 104 patients with cervical cancer were enrolled in this study. They received pelvic external beam radiotherapy and interstitial brachytherapy in five fractions. Both IPSA and manual optimized plan for isodose line were used to optimize the dose in each fraction. Dose volume parameters of the two plans were compared to analyze the dosimetric outcome by paired t-test. Results There were no significant differences in mean D 90and D 100for high-risk clinical target volume(HR-CTV)and D 90for intermediate-risk clinical target volume(IR-CTV)between the two groups(P>0.05). The IPSA group had a significantly higher D 100for IR-CTV than the manual optimized group(58.36±2.06 Gy vs. 53.99±2.17 Gy, P=0.025). For organs at risk,the IPSA group had a significantly lower mean rectum D 2ccand a significantly higher bladder D 2ccthan the manual optimized group(68.53± 2.85 Gy vs. 71.77± 1.79 Gy, P=0.002;80.49± 3.36 Gy vs. 78.71± 2.64 Gy,P=0.034). There was no significant difference in sigmoid D 2ccbetween the two groups(P>0.05). The IPSA group had significantly higher relative dose homogeneity index(HI)and conformity index (CI)of radiation dose for target volume than the manual optimized group(P<0.05), and there was no significant difference in overdose volume index(OI)between the two groups(P= 0. 1 0 7).Conclusions Compared with manual optimized plan for isodose line, IPSA can improve the dose distribution of tumor tissue,reduce mean rectum D 2cc,and increase CI and HI,so it is a preferable optimized treatment planning method in clinical application.

ABSTRACT

Objective To explore the clinical effect and influence on congnitive function of single use ofvalproate,oxcarbazepine,and levetiracetam in treatment of children epilepsy.Methods Children with epilepsy (96 cases) accepted in Weinan Matemty and Child Health Care Hospital from April 2011 to January 2016 were randomly divided into groups A,B,and C.Children in those three groups were given valproate,oxcarbazepine,and levetiracetam,respectively.Then the clincial efficacy,changes of EEG and congnitive function,and safety were compared.Results The total effective rates of groups A,B and C were 71.88%,78.13%,and 90.63%,respectively,group C was little higher than groups A and B without statistically significance.The epileptiform discharge improvement rate was getting better following extended therapy.The improvement rate of group C was better than that of group B and even much better than that of group A with statistically significance (P < 0.05).The verbal IQ (VIQ),performance IQ (PIQ),and full scale IQ (FIQ) of three groups before treatment had no big difference,but the scores of groups B and C were much better than before and group A after treatment with statistically significant difference (P < 0.05).The adverse reaction rates of groups A,B,and C were 28.13%,12.50%,and 3.13%,respectively with statistically significant difference among them (P < 0.05).Conclusion Valproate,oxcarbazepine,and levetiracetam have similar effect in treatment of children epilepsy,but oxcarbazepine and levetiracetam have positive significance in EGG and recognition function improvement.And the safety of levetiracetam is the highest of three drugs,which is worth of clincial application.

ABSTRACT

Objective To investigate the effect and influence on inflammatory factors of salbutamol aerosol combined with montelukast in treatment of cough variant asthma.Methods From Jan 2015 to Jan 2016,120 cases of cough variant asthma in our hospital were selected as the subjects.They were randomly and equally divided into 2 groups:control group (n =60) were treated with salbutamol aerosol,and observation group were treated with salbutamol aerosol and montelukast.The clinical effects,the pulmonary function and the serum inflammatory factors were compared between the two groups.Results After 2 months of treatment,the effective rate of the control group and the observation group were 73.3% and 96.7% respectively,there was statistical differences between two groups (P < 0.05).The FEV1 and FEV1/FVC of both groups showed remarkable increase.The FEV1,FEV1/FVC of observation group were (2.21 ± 0.17) L,(66.10 ± 4.46)%,and the FEV1,FEV1/FVC of control group (1.82 ± 0.11) L,(52.48 ± 3.34)%.The difference between two groups was statistically significant (P < 0.05).The IL-6 and TNF-α of both groups showed remarkable decrease.IL-6 and TNF-α in observation group were significantly lower than those of control group (14.58 ± 2.90 vs.20.87 ± 3.26,P < 0.05;0.59 ± 0.12 vs.0.97 ± 0.15,P < 0.05).Conclusion The use of montelukast combined with salbutamol aerosol in the treatment of cough variant asthma was safe and effective,and could significantly improve pulmonary function and reduce inflammatory factor.

ABSTRACT

Objective To investigate the genetic association between KCNJ10 rs1890532 gene polymorphism and susceptibility to children epilepsy in the Han descent population in the Northern China. Methods Two hundred twelve children with epilepsy were recruited as the epilepsy group. According to seizure types, they were divided?into partial seizures (PS) group (120 cases) and generalized seizures (GS) group (92 cases). Two hundred healthy volunteers without epilepsy were randomly selected from our hospital during the same period as the control group. General clini-cal data was collected and genotype was determined by polymerase chain reaction- restriction fragment length poly-morphism for the polymorphism of the KCNJ10 gene. Results Results showed that KCNJ10 rs1890532 gene polymor-phism was significantly associated with GS in children in both allelic association analysis (OR=1.325, 95%CI 1.014~2.461, P=0.043) and recessive model (OR=3.173, 95%CI 1.097~10.694, P=0.021) after adjustment for sex and age. There was no significant difference in any genetic model between the control group and epilepsy group, neither be-tween the control group and PS group. Conclusion Polymorphism of the KCNJ10 (rs1890532) gene may be associated with the risk of GS in children.

ABSTRACT

ObjectiveTo evaluate the application value of magnetic resonance diffusion weighted imaging (DWI)in diagnosis and differential diagnosis of prostate cancer. Methods53 Cases of suspected prostate cancer patients with elevated prostate-specific antigen, abnormaldigital rectal examination or ultrasound examination were underwent DWI examination before operation. Apparent dispersion coefficient was measured and compared to pathological results. Results There were 15 cases of prostate cancer, 37 cases of benign prostate hyperplasia and one prostatic intraepithelial neoplasia. When b =1 000 s/mm2 ,the ADC values of foci area in prostate cancer group and peripheral zone in benign prostate hyperplasia group were (0.813 ±0.25) × 10-3mm/8 and (1.45±0.24) × 10-3mm/s respectively, and the difference had statistical significance(t =-26. 117,P <0. 01 ). When b =60D s/mm2, the ADC values of foci area in prostate cancer group and peripheral zone in benign prostate hyperplasia group were ( 1.03 ± 0. 27) × 10-3 mm/s and ( 1.62 ± 0. 33) × 10- 3 mm/s respectively, and the difference had statistical significance( t =- 24. 191, P < 0. 01 ). The area under the ROC curves was 0.862, and the sensitivity and specificity of DWI were 78.1% and 91.5% respectively.ConclusionDWI can provide quantitative information for prostate cancer diagnosis and differential diagnosis,therefore it possesses a good clinical application value.
